Effect of Nicotinamide on Alcohol Metabolism



Abstract
An attempt was made to increase ethanol clearance for the treatment of acute alcohol intoxication by the administration of nicotinamide and the following results were obtained.
1. The administration of nicotinamide brought about a large increase of hepatic NAD in adult and lesser increase in young mice.
2. The NADH/NAD ratio was increased about two-fold in both of control and nicotinamide¡©treated mice by the administration of ethanol due to the reduction of NAD to NADH.
3. Alcohol dehydrogenase was mainly distributed in liver and increased with age reaching maximum one month after birth. The developmental change of hepatic alcohol lehydr¡©ogenase was closely related with ethanol metabolism, suggesting that ethanol met abolism in young mice is regulated by alcohol dehydrrogenase activity.
4. The increase of hepatic NAD after the administration of nicotinamide had no effect on the rate of ethanol clearance.
